Activity:
U0126 is a potent and selective non-competitive inhibitor of mitogen activated protein kinase (MAPK). U0126 inhibits MEK-1 and MEK-2 with IC50 values of 0.07 and 0.06 μM respectively, and has little or no effect on the activities of PKC, Abl, Raf, MEKK, ERK, JNK, MKK-3, MKK-4/SEK, MKK-6, Cdk2 or Cdk4. U0126 inhibits both the active and inactive forms of MEK-1/2. U0126 can enhance the self renewal of stem cells and it also helps maintain human pluripotent stem cells in an undifferentiated state. U0126 has an antiproliferative role in cancers, acting through different antitumour mechanisms in different tumours. U0126 is also able to potentiate or antagonise apoptosis, depending on the target cells.
